You're probably wondering exactly how Wes does simple western blots.
How Wes Performs Western Blots
The magic happens inside of a capillary. Each capillary cartridge can run either 13 or 25 capillaries — enabling throughput flexibility.
Running an immunoassay on Wes is simple.
Your sample, separation matrix, stacking matrix, antibodies, and reagents are loaded automatically from a specially designed plate.
Wes begins by aspirating the separation matrix and then the stacking matrix into each capillary, so proteins will stack tightly before they separate just like an sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE).
Next, your protein lysate is loaded.
Capillaries are lowered making contact with running buffer and voltage is applied to enable separation by molecular weight.
Once the separation is complete, UV light immobilizes the proteins to the capillary wall.
With proteins now immobilized and the matrices cleared of the capillary, Wes starts the immunoprobing by first incubating with your primary antibody of choice followed by a secondary HRP conjugate and finally chemiluminescent substrate.
The chemiluminescent reaction gives off light that is recorded by a CCD camera in a series of images over time with fresh substrate added before each exposure.
Three hours later, you'll get quantitative, size-based data.
