Simple Plex Assays for Viral Titer Quantification
Fast and reproducible viral capsid measurement is essential for those focused on gene transfer, as part of a cell and gene therapy workflow. While traditional plate-based and dot blot assays can be useful, they are often complex, time-consuming, and non-standardized.
Automated Simple Plex™ assays on the Ella™ platform offer a fast, sensitive, and reproducible method for reliable viral capsid measurement. Simple Plex assays are available to quantify the viral titer of both lentivirus and adeno-associated virus capsids.
- Measure viral titer across a broad dynamic range
- Get triplicate data results with just 25 µL or less of sample volume
- Streamline workflow with only 15 minutes of hands-on assay prep time.
- Achieve the same reproducible answers across multiple users and multiple sites
- Get highly reproducible validated data with no manual steps in just 90 minutes

FAST AND REPRODUCIBLE AAV2 VECTOR TITRATION
To meet increased throughput and safety demands, investigators are seeking more automated analytical solutions for assessment of viral vector identity, purity, and stability. The Simple Plex AAV2 assay on the Ella platform provides an accurate and efficient method for measuring the AAV2 capsid titer. The assay uses the recombinant AAV2 antibody (A20R) from industry leader PROGEN to quantify assembled AAV2 capsids.
Combining the proven specificity of PROGEN AAV2 antibodies with the convenient workflow and robustness of the Ella platform, the Simple Plex AAV2 viral titer assay:
- Offers a broad viral detection range combined with hands-free automation to accelerate AAV2 viral capsid quantification
- Uses factory calibrated assay cartridges, which help minimizes setup time and reduce user-dependent variability while reducing the need for sample retests
- Relies on the innovative Ella platform to perform every step of the immunoassay automatically, from wash steps to final
- Provides a robust alternative to the conventional ELISA or dot blot approaches
- Delivers superior assay sensitivity, helping to ensure data quality and consistency
Comparing the Simple Plex AAV2 Assay with the PROGEN AAV2 ELISA
In this application note, we demonstrate assay comparability between the Simple Plex AAV2 assay and the PROGEN AAV2 Titration ELISA (PRATV) to show that you get the same high-quality data you would expect from the PROGEN ELISA with an automated, streamlined workflow.

With the growing use of lentiviral vectors derived from HIV-1 in gene transfer workflows, researchers need fast, reliable viral titer quantification during process development. The 3-to-6-hour timeframe required to run a standard ELISA is a challenge for operators who increasingly require results quickly to inform real-time decision making.
The Simple Plex assay for the detection of human HIV-1 Gag p24 combines high throughput with superior accuracy and precision to enable rapid quantification of lentiviral capsid proteins throughout different stages of cell and vector manufacturing. The automated, reproducible nature of the Simple Plex p24 assay helps eliminate user error, making it an ideal tool in good manufacturing practice (GMP) environments where quality, standardization of process, assay transfer, and scalability are highly desirable.

What are viral titer assays?
Viral titer assays are designed to quantify viral vector concentration. In addition to cell and gene therapy gene transfer workflows, viral titer assays are integral to the study of infectious disease pathogenesis and vaccine development. At a high level, viral titer assays quantify: DNA titer, infectious titer, or capsid titer. DNA titer assays quantify the number of viral capsids containing DNA. Infectious titer assays quantify infectious particles that transfer DNA into host cells via a process called viral transduction. Capsid titer assays quantify viral capsids. Capsids may or may not contain viral DNA.
