B7-H1/PD-L1 (programmed cell death 1 - ligand 1; CD274) is a significant cancer immunotherapy target of interest. It is a potential predictive cancer biomarker whose overexpression on tumor cells suppresses the immune response against the tumor. The PD-1 receptor (programmed cell death 1; CD279), and its ligand PD-L1 belong to the family of immune checkpoint proteins that act as co-inhibitory factors, and can inhibit the T cell response.
PD-1 is expressed on the surface of activated T cells while PD-L1 is commonly expressed on the surface of dendritic cells, macrophages, and overexpressed on tumor cells for many cancers. Immunoassays enable the measurement of PD-L1 overexpression despite some known limitations. For example, detection of PD-L1 via immunohistochemistry (IHC) often produces variable results. Other methods like manual ELISAs lack low pg/mL sensitivity and are tedious to setup and run, making them more error-prone.
